Identification of the A3 adenosine receptor in rat retinal ganglion cells.

چکیده
PURPOSE Adenosine can protect retinal ganglion cells from the death that accompanies a general ischemic challenge as well as excitotoxic death. In other tissues, both A1 and A3 adenosine receptor subtypes can mediate protection. While a role for the A1 adenosine receptor in ganglion cell protection has been established, a potential for the A3 receptor has only recently been proposed. Although the pharmacology is promising, the molecular identity of the responsible receptor is unclear as previous studies were unable to detect message for the A3 receptor in retinal ganglion cells. We combined laser capture microdisection (LCM) and immunopurification with traditional and real-time PCR to unequivocally demonstrate the presence of the A3 receptor message in rat retinal ganglion cells. METHODS Retinal ganglion cells of Long-Evans rat pups were retrograde labeled with aminostilbamidine. Eyeballs were enucleated, embedded, frozen, sectioned, and fluorescent cells in the ganglion cell layer were collected with LCM. Purified ganglion cells were also isolated with a two-step panning procedure. cDNA for the A3 receptor obtained from the microdissected ganglion cell layer, immunopurified ganglion cells, whole retina and testis was amplified using RT-PCR, confirmed by DNA sequencing and compared with published sequences. A3 receptor message was also amplified using real-time PCR. Ca2+ levels in immunopanned ganglion cells were measured ratiometrically with fura-2. RESULTS RNA from immunopurified ganglion cells and from dye-loaded cells in the ganglion cell layer contained message for the A3 receptor when amplified with either traditional RT-PCR or real-time PCR. The entire encoding region was sequenced and found to be 99% identical to the published code. The sequence closely resembled the consensus form of the gene, with other sequences deviating from this default code. Molecular identification was functionally confirmed in purified ganglion cells as the A3 receptor agonist Cl-IB-MECA prevented the excessive Ca2+ rise triggered by P2X7 agonist BzATP. CONCLUSIONS Retinal ganglion cells express A3 adenosine receptor mRNA. Stimulation of this receptor can reduce the Ca2+ overload following excessive activation of P2X7 receptors.

Selective Inhibitory Effect of Adenosine A1 Receptor Agonists on the Proliferation of Human Tumor Cell Lines
Background: In this study, the effects of three structural analogues of adenosine upon proliferation of human tumor cells were investigated. Previous research showed a cytotoxic effect of adenosine via A3 receptor and A1 receptor and sometimes this effect was receptor independent. The researches showed a differential cytotoxic effect of adenosine and its A3 agonists on cancerous cells, while ot...
متن کاملThe role of adenosine A3 receptors in cytotoxicity of the breast cancer cell lines
The nucleoside adenosine is present within cells and body fluids of all living organisms and its production, both intra- and extracellularly, is tightly coupled to energy consumption resulting in increased level of extracellular adenosine. The physiological effects of adenosine are mediated through four pharmacologically and biochemically distinct adenosine receptors (AR), i.e. A1, A2A, A2B and...
متن کاملAdenosine-Evoked Hyperpolarization of Retinal Ganglion Cells Is Mediated by G-Protein-Coupled Inwardly Rectifying K and Small Conductance Ca -Activated K Channel Activation
Adenosine is a neuromodulator that activates presynaptic receptors to regulate synaptic transmission and postsynaptic receptors to hyperpolarize neurons. Here, we report that adenosine-induced hyperpolarization of retinal ganglion cells is produced by the activation of A1 receptors, which initiates a signaling cascade that activates G-protein-coupled inwardly rectifying K (GIRK) channels and sm...

Glaucoma is the leading cause of preventable blindness worldwide. Despite tremendous advances in medical and surgical management of glaucoma in the recent years, the prevalence of glaucoma related blindness is anticipated to increase in the future decades because of the aging population. Stem cells have the potential to change the glaucoma management in several ways. There are several areas of ...
متن کاملChloride channel protein 2 prevents glutamate-induced apoptosis in retinal ganglion cells
Objective(s): The purpose of this study was to investigate the role of chloride channel protein 2 (ClC-2) in glutamate-induced apoptosis in the retinal ganglion cell line (RGC-5). Materials and Methods: RGC-5 cells were treated with 1 mM glutamate for 24 hr. The expression of ClC-2, Bax, and Bcl-2 was detected by western blot analysis. Cell survival and apoptosis were measured by 3-(4,5-dimeth...
